The significance of Harry's eyes are on Snape. Snape was... still is, I think, in love with Lily. Harry's eyes (a carbon copy of Lily's) remind Snape of Lily, that's why he looked into Harry's eyes before he died.
The 'gleam of truimph' in Dumbledore's eyes happened because upon knowing of Voldemort taking Harry's blood into himself, meant that the protection still lives and that Harry can't die because of it.

Snape loved Lily, and Harry had his mother's eyes. When Snape died he said "look...at...me" and Harry looked, and the last thing Snape saw was Lily's eyes. There was SO much eye contact between Harry and Snape in the books (besides the fact we knew Snape could look into Harry's mind.) But he hated Harry because he looked like his Dad, but had his mother's eyes.
The gleam was that he knew that Harry would not die while Harry's blood ran through Voldemort's vein. He knew Harry would be triumphant.
